Ultrathin sectioning

The inner condition of TEM has to keep high vacuum to transmit electron beam. It is impossible to observe the native sample, so it has been in progress about sample preparation. The ultrathin cross sectioning technique was generally used and the procedure was followed: 1, sample was fixed with chemicals, 2, dehydration in alcohol of elevated concentration, 3, the sample was embedded by resin and 4, hardened sample was cut ultrathin section around 60nm using diamond knife.
